C8 packing material is a reversed-phase stationary phase with octyl ligands bonded to silica. Compared with a more hydrophobic C18 phase, C8 may be considered when a process requires lower hydrophobic retention or an alternative selectivity profile. The correct stationary phase must be confirmed through method development using the actual sample, mobile phase, loading, target purity, and throughput requirement.
HP-C8 is supplied as bulk chromatography packing material for column packing. Haipu can also provide custom pre-packed preparative C8 columns based on the requested column dimensions, application, and packing requirement. HP-C8 can be used with standard reversed-phase preparative chromatography equipment, including dynamic axial compression columns. For a compact packed bed, use approximately 1.05–1.10 times the calculated packing requirement.
Methanol, isopropanol, or ethanol can be used as slurry solvents. A slurry concentration of 30–40% (w/v) is a recommended starting point. For DAC packing, the reference packing pressure is 80–100 bar (8–10 MPa) for 10 μm silica packing. Preparative operating pressure should remain below the packing pressure.
After packing, evaluate theoretical plate number and peak symmetry using conditions appropriate to the process. Begin operation at a low flow rate and increase gradually after the column pressure and baseline are stable.
Operate HP-C8 within 10–60°C and within the stated pH stability range. Filter feed solutions and mobile phases through 0.45 μm filtration where appropriate; pre-treat complex feeds or use a guard column when necessary.
After use, first remove salts with a low-organic mobile phase when applicable, then clean with a stronger organic solvent. Methanol, acetonitrile, isopropanol, or a 95% dichloromethane / 5% methanol mixture may be considered according to process compatibility. Do not use nitric acid to clean HP-C8 packing material.
Store unused material sealed in its original container in a cool, dry location at 4–25°C. The typical shelf life is two years when stored as supplied.
