HP-Wang Resin

Wang resin is currently the most widely used solid-phase peptide synthesis resin, which can be used for the immobilization of carboxyl and phenolic hydroxyl groups in solid-phase synthesis. After ester bond formation, it maintains good stability under various reaction conditions but can also be easily removed from the resin by moderate acid treatment, usually trifluoroacetic acid. For Wang resin immobilizing amines, it can also be rapidly converted into an equimolar urethane-protected solid-phase support by reaction with phosgene or active carbonates (such as N,N'-carbonyldiimidazole or bis(p-nitrophenyl) carbonate).

Characteristics

The most widely used solid-phase support for acidic substrates. The functional group is a hydroxyl group, and the first amino acid is linked to the resin via an ester bond under DMAP catalysis. Its linker is relatively stable and requires cleavage with high-concentration TFA.

Applications

Suitable for the solid-phase synthesis of most peptide acids. However, measures should be taken to avoid racemization when the first linked amino acid is Cys or His, and attention should be paid to preventing DKP side reactions when the C-terminal dipeptide contains Gly or Pro.
